5 feb. 2020 — Kozarcanin, H., Lood, C., Munthe-Fog, L., Sandholm, K., Hamad, O.A., et al. (2016). The lectin complement pathway serine proteases (MASPs)
The complement system is activated cascadically via three distinct major routes: classical pathway (CP), alternative pathway (AP) or lectin pathway (LP). The unique factors associated with the latter are collectins (mannose-binding lectin, collectin-10, collectin-11), ficolins (ficolin-1, ficolin-2, ficolin-3) and proteins of the mannose-binding lectin-associated serine protease (MASP) family
This occurs through MBL pathway which is activated when circulating lectin (MBL) binds to mannose residues on glycoproteins or carbohydrates on the surface of microorganisms. The lectin complement pathway is initiated by microorganism-bound MBL as it associates with two human MBL-associated serine proteases (MASP-1 and -2) that cleave and activate C4 and C2. MASP-1 and MASP-2 are similar to C1r and C1s, respectively, and MBL is similar to C1q. The lectin pathway activates C3 convertase by using a Lectin. Lectin is a special protein that binds to specific carbohydrates. When macrophages ingest viruses, bacteria and other foreign material they release the cytokines. These cytokines stimulate liver cells to secrete mannose-binding protein (MBP), a lectin for mannose.

Lectin Pathway Activation Human ficolins activate the lectin complement pathway. In human serum, ficolin-2 and ficolin-3 form complexes with all three types of mannose-binding lectin (MBL)-associated serine proteases (MASP) and their truncated proteins, sMAP (MAp19) and MAp44 (MAP-1). Discrimination between self and non-self by lectins (carbohydrate-binding proteins) is a strategy of innate immunity that is found in both vertebrates and invertebrates. Se hela listan på academic.oup.com
Here we investigated the role of a central humoral component of innate immunity, the lectin pathway of complement, in recognition of mitochondria in vitro and in vivo. We found that the soluble pattern recognition molecules, mannan-binding lectin (MBL), L-ficolin, and M-ficolin, were able to recognize mitochondria. C3−/−, MBL1 Objective. The pathogenesis of systemic lupus erythematosus (SLE) involves complement activation. Activation of complement through the classical pathway (CP) is well established. However, complement activation through pattern recognition not only happens through the CP, but also through the lectin pathway (LP). We investigated the hypothesis that the LP is activated in SLE and involved in the 2002-05-01 · The lectin-complement pathway--its role in innate immunity and evolution.

Mannose-binding lectin (MBL), also known as mannan-binding protein (MBP), is a protein that is involved in complement activation via the lectin pathway. The complement system provides immediate defense against infection and has proinflammatory effects. MBL deficiency has been variously defined as a serum levels either <100 or <500 ng/mL.
